Turn raw assay videos and plate exports into clean curves and figures in 3 days.
Hand-scoring wells and fitting curves in spreadsheets misses uneven lighting, block temperature gradients, neighbor heat carryover, and frost halos. DarkBiome Daybreak pulls calibrated reaction rates, melting temperatures, and freezing curves directly from raw videos, microscope stacks, and plate-reader files.
One pipeline for melting curves, clearing videos, thermal cameras, and freezing runs.
Validated across 27 public benchmark datasets (41,800 video and multi-page TIFF frames plus 6,964 plate-reader melt curves), Daybreak processes enzyme assays, stability screens, calorimetry videos, and cold-stage experiments without custom lab code.
Melting Curves & Thermal Shift
Standard plate-reader DSF exports (CSV, LightCycler, qPCR melt, Excel), microwell fluorogenic melts, 48–96-droplet optical ramps, and live-cell thermal aggregation (RT-CETSA) curves with clean Tm and Tagg fits.
Clearing, Fluorescence & Lysis Videos
Polymer-clearing assays (polyurethane and BHET polyester suspensions), 16-bit fluorescence microscope TIFF stacks, flatbed scanner growth plates, colorimetric turnover, fibrin clot lysis, mineral precipitation, and droplet cell lysis.
Thermal-Camera Reaction Heat
Infrared recordings of 96-well plates and microfluidic channels (FLIR, InfraTec, Optris, Telops, Seek), separating real enzyme or catalyst heat release from neighbor-well thermal carryover and lighting artifacts.
Droplet & Plate Freezing
96-well and dual-96 plates, PDMS microwells, ungridded droplet slides, and cryo-microscopy ice fronts—automatically flagging neighbor frost-halo cascades and correcting for block gradients and solute depression.
01 · You Provide
- Raw camera video (MP4/AVI), multi-page microscope/scanner TIFF stack, or infrared thermal video (96-well, dual-96, PDMS chip, or microfluidic array; 384-well not offered).
- Or raw 96-well fluorescence, absorbance, or luminescence plate-reader export (CSV, XLSX, RDML, or LightCycler).
- Well or channel map (sample IDs, concentrations, volumes, buffer composition, and temperature probe log if heated or cooled).
02 · What We Run
- Well & droplet tracking: tracks brightness, turbidity, fluorescence, or temperature per well while subtracting uneven lighting, neighbor heat bleed, and frost-halo cascades.
- Curve & rate fitting: extracts melting temperatures (Tm / Tagg), initial reaction and clearing slopes, integrated reaction heat, or corrected freezing spectra.
- Calibrated error bars: folds probe accuracy, edge-to-center block temperature offsets, and ramp lag into 95% confidence intervals per sample.
03 · Deliverable Package
- Annotated check video or image overlay marking every tracked well, inflection point, thermal peak, or excluded neighbor artifact.
- Publication-ready SVG/PDF figures + clean CSV and Excel summary tables ready for supplementary data.
